1. Microbes - the source of biological products
Microorganisms are rich resources for various biologically active products, regardless of past, present and future. There are many biologically active substances, such as primary metabolites of microorganisms, such as amino acids and vitamins; and secondary metabolites of microorganisms, such as antibiotics. The key to obtaining a new product of the desired biological properties is the source of the biological product---the choice of microorganisms; what screening scheme (detection system) is employed. For the past 40 years, the focus has been on screening antibiotics for medical use.
Antibacterial treatment: penicillin, cephalosporin and tetracycline antibiotics; anticancer and antifungal infections: mitomycin, griseofulvin, etc.; other treatments: hypertension drugs, blood cholesterol steroids ( Hypocholesteremic agents) and immunomodulators (immunomdulator).
Due to the complexity of the microbial cell's inclusions and the surrounding medium composition, and the desired product may be only an order of magnitude from picogram to milligram per milliliter, no matter which type of biologically active substance is sought, it must be selected when selecting a screening method. Considering both selectivity and sensitivity, a highly sensitive method of specificity detection is required.
2. The nature of the sample to be screened
Producing bacteria for new products can be screened using solid and liquid media. However, the large-scale production of secondary metabolites is carried out by submerged culture method. Because of the different ways of producing secondary metabolites in solid medium and liquid medium, some manufacturers use liquid culture method as the only screening method. .
3. Design of screening scheme
Basically, three different screening methods can be utilized: 1 overall organism; 2 intact cells; 3 subcellular preparations. The first is the most straightforward method of drying active substances in the body, but considering the small number of samples that can be processed, the cost is high and the screening method is not sensitive enough. It is not appropriate to use this method as the initial screening.
