According to the theory of "corresponding to heaven and man" and the theory of yin and yang, the Chinese medicine doctor Wang Qiaochu prepared peanut stalks with the characteristics of "opening the night" into a single preparation "891" Anshen series for the treatment of insomnia and achieved good curative effect. In this experiment, the spontaneous activity of mice and the synergistic effect with sodium pentobarbital were detected by photoelectric method. The sedative and hypnotic effects of "891" Anshen I and II were studied.
1 Materials and methods
111 test drug Zaoren Anshen oral liquid, Beijing Tongrentang Pharmaceutical Factory (batch number: 960107); Anding injection, Xudong Haipu Pharmaceutical Co., Ltd. (batch number: 960416); peanut branch extract B, provided by the Department of Chemistry, Shanghai Jiaotong University; "891" Anshen I (batch number: 960802), II (batch number: 970301) and peanut branch extract H, Shanghai Huangshan Pharmaceutical Factory, all doses are converted into raw dose.
112 Animals Kunming mice, clean grade, male, weighing 18-22 g, were provided by the Experimental Animal Department of Fudan University Medical College. Before the experiment, the animals were fasted for overnight.

1311 Spontaneous activity of mice (photoelectric method) Animal selection criteria were determined before the experiment: 10 mice were randomly selected and placed in the test box, and the number of blocked light was recorded in 15 minutes, and the number of animals covered in the upper and lower limits was taken as the range of 100. The most numerical values ​​are the screening criteria. Each mouse was screened before the experiment. Those who passed the test were placed in the test box according to the group. After 5 minutes, the instrument was turned on to count and print, and the light was 0~, 16~, 36~, 46~60 minutes. The number of blocked times is the detection result.
11312 Synergistic effect with sodium pentobarbital Sodium pentobarbital solution was intraperitoneally injected 15 minutes after intragastric administration. In the experiment of prolonging the hypnotic effect of sodium pentobarbital, the dose of sleeping dose was 64 mg/kg, which was the index of disappearance of righting reflex in mice; the subthreshold dose of 54 mg/kg in the experiment of intensive hypnosis was small. When the mouse righting reflex disappeared for more than 1 minute, it was recorded as sleep, and the percentage of sleep in each group of mice was counted.

Effect of 212 “891” Anshen I and Extract H on spontaneous activity in mice
The effect of "891" Anshen I and the spontaneous activity of the P-parts of Peanut branch in Table 1 was basically the same, and both of them were significantly reduced; Extract H also had a strong effect. Adding H to "891" Anshen I can improve its sedative effect, and there are significant differences in the 16-time period, and there is also a good dose-effect relationship.
213 Synergistic effect of "891" Anshen Mixture II and pentobarbital sodium
50g/kg of mixture II can prolong sleep time, significantly increase sleep rate (%), sleep time (min), control is 3012±2710, II is 5713±3616 (P > 0.05); sleep rate control group 0, II is 60% (P < 0.01).
3 Discussion
Since the end of the 1980s, we have carried out a series of clinical and sedative effects on the sedative and hypnotic effects of yarrow flowers, acacia leaves and peanut leaves with the characteristics of "opening the night" according to the "correspondence between heaven and human" and the theory of yin and yang. Experimental studies have found that they have a good effect. On the basis of the preliminary clinical proven efficacy of peanut branches and leaves, this experiment was provided by Professor Du Shangjian from the Department of Chemistry of Shanghai Jiaotong University to find the effective part of the sedative and hypnotic effects of peanut leaves. After repeated experiments, it was determined that extract B had a very significant effect. Based on this, we determined the process of producing “891” Anshen I from single-flavored peanut leaf extract B, which was produced in batches by Shanghai Huangshan Pharmaceutical Factory and used in clinical practice. The preparation has good sedative effect (see Table 2); The extract H obtained in the production process has been confirmed by experiments to inhibit the spontaneous activity of mice. The combination with "891" Anshen I can further improve the sedative effect. The production process of "891" Anshen Mixture II was designed, and the pharmacological proof proved to have a good hypnotic synergistic effect with sodium pentobarbital, and satisfactory results were obtained in clinical practice. The preparation was recently approved by the Shanghai Science and Technology Commission, with the participation of Shanghai Mental Health and Huashan Hospital in a double-center, randomized, double-blind, placebo-controlled clinical trial. The total effective rate of the “891” Anshen Mixture II group was 7316%. The placebo group was 4312%. The results suggest that there is efficacy and no toxic side effects have been found.
