BEIJING, Beijing, March 8 (Song Dongzhuang Yingna) Chinese scientists completed the first molecular proteomic typing of tumors in the world. The latest results show that diffuse gastric cancer can be divided into three subtypes at the proteome level, and the subtypes of different subtypes are diagnosed by the Institute of Military Medical Sciences of the Chinese People's Liberation Army Academy of Military Sciences and Peking University Cancer Hospital. Different, the sensitivity to chemotherapy is also different.
On March 8th, Beijing time, the internationally renowned academic journal "Nature? News" published the research results online.
Qin Yu, director of the National Institute of Protein Science (Beijing), said that this protein typing does not depend on any genomic information, reflecting the importance of the information provided by the protein as a direct performer in clinical science. Comprehensiveness can provide a direct basis for accurate medical care for patients with gastric cancer.
Gastric cancer ranks third among global causes of cancer death. The data show that the number of new cases of gastric cancer in China is about 680,000 per year, accounting for about half of the global cases. At present, the widely used Lauren classification morphologically divides gastric cancer into intestinal type, diffuse type and mixed type. Among them, diffuse type gastric cancer is called "stomach cancer in gastric cancer", and there is no effective targeted therapy at present. Previously, molecular typing of gastric cancer based on genome and transcriptome was published internationally, but no effective association with clinical prognosis was achieved.
In 2012, Qin Hao led the team to open the first diffuse gastric cancer proteomics map and tumor-driven genetic variation map. At the same time as Juli opened the technical route of gastric proteomics analysis, they began a large-scale analysis of gastric cancer samples in conjunction with Peking University Cancer Hospital.
In June 2016, the research team completed 84 mass spectrometry measurements of gastric cancer and paracancerous tissue samples, and identified 11,340 different gene products. These proteins cover all possible proteins expressed in tumor cells and tumor microenvironments, thus for the first time depicting the proteomic full spectrum of diffuse gastric cancer.
Further studies have found that only 84 pairs of gastric cancer and paracancerous tissue protein expression profiling cluster analysis, diffuse gastric cancer can be divided into three molecular subtypes closely related to survival prognosis and chemotherapy sensitivity. On this basis, the research team also screened 23 prognostic-related gastric cancer candidate protein drug targets.
According to Qin said, gene sequencing can only provide very limited clues for accurate medical treatment of tumors. This confusion can now be solved at the protein level. "In the future, in the clinical, proteome analysis of tumors, you can know which kind of tumor, whether it can be cured, which treatment should be taken, and which treatment should not be taken.
