Hybridoma technology is still the main technology for the discovery of monoclonal antibodies, especially in the field of therapeutic antibody therapy, compared with non-natural libraries such as phage and yeast, which have the characteristics of good drug-making and strong specificity. Since the monoclonal antibody obtained by the hybridoma is a sequence obtained by natural evolutionary screening of mammalian cells, it usually does not encounter problems such as difficulty in expression, low yield, and non-specific binding.
Hybridoma technology has the following challenges:
1. For antigens with low immunogenicity, antibody titers are low and it is difficult to produce neutralizing antibodies;
2. The fusion is inefficient and it is difficult to obtain clones with low abundance;
3. Low antibody affinity and epitope distribution limit.
In response to the above limitations, Qiwen Bio has proposed the following optimization schemes:
1. For low immunogenic antigens, immune-enhancing adjuvants increase antibody titers
METHODS: Our immunoenhancing adjuvants extend the epitope recognition of T cells based on traditional adjuvants such as CFA, thereby expanding the epitope of antibodies.
RESULTS: Multiple trials showed that after initial immunization, the titer could be increased by 5-10 times.
2. Positive clone adjuvant enhances B cell abundance in response to the low positive rate of fusion
METHODS: In the Pre-fusion boost phase, our positive clone adjuvant enhances the abundance of positive clones in B cells and helps to fuse to rare clones by increasing the positive rate.
RESULTS: Multiple trials have shown that for most antigens, the modified method can increase the positive rate by 3 to 5 times.
3. Specialized culture system to improve integration efficiency
METHODS: We provided domestically-preserved mouse SP2/0 cells and hybridoma-specific culture sera. For the combination of advanced electrofusion technology, the experimental effect of our self-produced electrofusion buffer is comparable to that of imported products.
Result: According to customer feedback, the fusion efficiency can be up to 1:1000, usually 1:5000 or above. If used in combination with the fusion treatment solution, the fusion efficiency can be further improved (the fusion efficiency is increased by 3 to 5 times).
4. Hybridoma cell enhanced medium increases colony growth rate
Methods: Hybridoma cell-enhancing medium was provided during the clonal screening and growth stages following hybridization of hybridoma cells. The medium contains a variety of cytokines that promote the growth of hybridoma cells.
Results: The clonal growth rate can be increased by nearly 10 times.
