Introduction to melamine

In September 2008, the outbreak of Sanlu infant milk powder in China caused a kidney stone disease in infants and young children who consumed contaminated milk powder because the milk powder contained melamine. The General Administration of Quality Supervision, Inspection and Quarantine (AQSIQ) notified the results of the melamine content of infant milk powder in the country, and detected melamine in 69 batches of products from 22 manufacturers including Hebei Sanlu, Shanxi Yashili, Inner Mongolia Yili and Mengniu Group, and was required to immediately remove the melamine. China Central Television carried a report in the news network's program 19:00. The incident has attracted widespread attention in China and has also hit China's manufacturing commodity reputation. Many countries have banned the import of Chinese dairy products.

So what is melamine? Why is melamine added in dairy products? Melamine, commonly known as melamine and protein essence, is a triazine-based nitrogen-containing heterocyclic organic compound. It is used as a chemical raw material. Because its chemical structure contains many "amino" nitrogens, the nitrogen content is quite high, so it is often added by illegal traders. In the food, the content of the "protein" of the food is increased, and the melamine is made of urea as the raw material, and the production price is low, which is also the main reason for the illegal addition of melamine as an additive.

The addition of melamine to foods can cause great damage to our body, especially in the kidneys. Animal toxicology experiments have shown that taking melamine causes a large amount of white crystals to precipitate in the kidneys and ureters, causing kidney swelling, bleeding and even exhaustion. Some will also form stones, block the urinary system, causing greater damage. In this regard, relevant national authorities have established relevant residue limits to ensure food residue safety.

Residue limit:

China National Food Quality Supervision and Inspection Center pointed out on September 13 that melamine is a chemical raw material and is not allowed to be added to food. Therefore, standard restrictions such as pesticide residues have not been set. On October 8 , the Ministry of Health, the Ministry of Industry and Information Technology, the Ministry of Agriculture, the State Administration for Industry and Commerce and the General Administration of Quality Supervision, Inspection and Quarantine jointly issued an announcement to establish the temporary management values ​​of melamine in milk and dairy products:

The limit of melamine in infant formula milk powder is 1mg/kg , and products above 1mg/kg are not allowed to be sold. The limit of melamine in liquid milk (including raw milk), milk powder and other formula milk powder is 2.5mg/kg , and products above 2.5mg/kg are not allowed to be sold. The limit of melamine in other foods containing more than 15% milk is 2.5mg/kg , and products above 2.5mg/kg are not allowed to be sold.

The melamine in the sample competes with the antigen-specific antibody immobilized on the plate, and the enzyme standard II antibody is added to catalyze color development, and the content of melamine in the sample is judged according to the depth of color development. The color is deep, the content is small, the color is light, and the content is high.

Read the instructions carefully to prepare the reagents (such as washing solutions) required during the experiment.
Sample preparation:
1. Take out the raw milk of the sample to be inspected from the refrigerator;
2. Take 1 mL of the original milk sample that is equilibrated to room temperature and mix well in a 2 mL centrifuge tube for labeling.

1. Remove the unneeded micropores into an aluminum foil pouch and store in a plastic pouch.
2. Prepare the required reagents, microplates, micropipettes and tips, and start the plate operation.
3. The standard working fluid/sample, the enzyme standard II anti-working fluid, and the antibody working fluid are sequentially added to the micro-pore of the enzyme label according to the instructions, and timed.
4. After the reaction time is completed, the liquid in the enzyme standard plate is thrown away, the plate is washed four times in succession, and patted dry;
5. After mixing the AB liquid 1:1, add 100 μL to each well, and time for 15 minutes for color development;
6. After the color development time was reached, 50 μL of stop solution was added to each well and data was read within 5 min.
Fifth, data processing analysis
1. Open the Widwekang analysis software and read the data;
2. Perform data setting of standard products and samples in the interface of the software;
3. After the setting is completed, set the layout on the interface of the analysis software, click the data analysis button, and print the experiment report or store the data and report according to the requirements.
